How to Perform a DNA Sample Collection for DNA Testing

DNA is the hereditary material or genetic instructions in humans and other organisms. Almost all cells in the body have the same DNA. DNA is located mainly in the cell nucleus but a small amount is located in the mitochondria. DNA extractions are from skin, muscle, bone marrow, saliva and cheek cells. DNA is used in solving crimes and for paternity testing.

Instructions

Step 1
Identify all items found inside your free kit for the collection of DNA for testing. Detailed instructions explaining the collection process along with 4 buccal swabs, 4 buccal swab envelopes to hold the samples and a business reply envelope for shipment the samples.
Step 2
Label the buccal swab envelope where indicated with the required information.
Step 3
Swab the inside of your cheek with the buccal swab provided in the kit. Use one of the swabs to rub against the inside of your cheek lining, rotate the swab while covering as much of the cheek area as possible.
Step 4
Put the used swab in pre-labeled envelope. Do not use the plastic wrapper that enclosed the swab; this can cause growing molds which will destroy the DNA sample.
Step 5
Repeat Step 3 and 4 with another swab for the same cheek. Following the same procedure use the remaining two swabs for the other cheek and seal the envelope with all 4 used swabs.
Step 6
Place the sealed envelope into the kit, put the entire collection kit in the business reply envelope provided and mail this package back to the laboratory for testing.
Step 7
Results from the paternity test should be available within 5-7 business days.

Where are samples collected?

We will provide you, or your client, with the location (usually a local laboratory or clinic), date, time for sample collection, and the name of a contact person at the site.

Are there any age requirements?
Paternity testing using DNA testing may be performed at any age. Age of the person(s) tested DOES NOT affect the accuracy of the paternity test / DNA test result in any way. We have successfully performed numerous tests involving newborn infants.

Who needs to provide a sample?
Usually, a sample is collected from the mother, child and alleged father. Under special circumstances, DNA testing / paternity testing may be performed using only samples from the child and alleged father. Mother sample can be optional.

What types of samples are collected?
In most cases DNA testing / paternity testing is done using cells from the inside of the mouth, which are collected with a cotton swab called a "buccal swab". The older more classic approach is to collect a very small sample of blood. The sample is small enough to allow even newborns to participate in testing. Newborns may also be tested using umbilical cord blood obtained at birth. Relationship testing can be performed on various discreet samples like Toothbrush, Hair sample, used bandage, contraceptives, nail clippings or any other samples where possibility of human dna exist. Please note that, our lab make best efforts to get good yield of DNA from these sample but some times it fails due to non sufficiency of DNA.

Legal DNA Paternity Test in India

The type of paternity test you need will depend on what you intend to use the DNA test results for:
  • If you need paternity test results that can be used as a legal document (for example, to change the name on the birth certificate or to obtain child support and other benefits), a Legal DNA Test needs to be performed (described below).
  • However, if you need the test only for personal knowledge, a Home DNA Test willl suffice.
Unlike the Home DNA Test, where tested parties collect their own samples at their convenience, the Legal DNA Test follows a Chain of Custody documentation process to ensure that you receive accurate and legally defensible results. DNA Test Participants
In a standard DNA paternity test, the tested parties include a child, the alleged father, and the mother (called a trio).
The mother’s participation in the paternity test helps to exclude half of the child’s DNA, leaving the other half for comparison with the alleged father’s DNA. However, we can perform a paternity test without mother’s participation (called a motherless). A motherless test involves additional analysis, which we performs without any additional charge. Results are equally conclusive whether or not the mother participates. Motherless tests are guaranteed to have at least a 99.9% probability of paternity for inclusions and 100% for exclusion.

A Twin Zygosity Test is a DNA test that definitively shows whether twins are identical or fraternal.

During OB-GYN visits, the physician might be able to tell whether twins are identical or fraternal through ultrasound examination of the placenta. When the twins are born, pathological examination of the placenta can also be done to determine zgosity.

However, studies have shown that either method is not 100% accurate, and scientists recommend DNA testing to determine zygosity. There are also many cases in which medical records regarding zygosity have been lost, or doubt may have arisen because of the twins’ physical characteristics as the grow up.

In such cases, only a DNA test will be able to reveal the truth. A twin zygosity test compares the twins’ DNA profiles to see whether they match—an exact match proves that the twins are identical.

In situations where the alleged father is unavailable for testing such as he is deceased or unwilling to participate, the alleged father's parents can be tested to determine whether they are the true biological grandparents of the child. When conducting a grandparentage test, it is recommended to test both grandparents (the grandmother and the grandfather) in order to obtain the most conclusive results.
If this is not possible, a grandparentage test can be conducted by testing only one grandparent. The sample of a common parent (normally the Mother) is also recommended to strengthen the results of the test.

A sibling ship test is a DNA test conducted to determine if two children share one or both parents (i.e., if they are half or full siblings). It is an indirect way to determine family relationships when an alleged father is not available for a paternity test.

A sibling ship test starts with the analysis of known relatives:

Sibling 1 and Sibling 2 do not share the same mother and they want to find out if they share the same biological father—in this situation, a half siblingship test is performed.

Sibling 1 and Sibling 2 share the same biological mother but are unsure if they share the same biological father—in this situation, a full siblingship test is performed.

In the test, the siblings’ DNA profiles are compared to see how much of their DNA could have come from a common father. Participation of the mother(s) is encouraged to help exclude the mother’s contribution to the children’s DNA. Siblingship tests require more analysis, and they could be more costly and take a longer time to complete without the mother’s participation.

Prenatal Paternity Test in India

Patients who wish to determine paternity during pregnancy may take a prenatal paternity test. Like a standard paternity test, the prenatal test compares the baby’s DNA profile with that of the alleged father. Because the baby’s DNA is set at conception, the prenatal test gives the same conclusive results as the standard paternity test.
To obtain samples from the unborn child, an OB-GYN uses either of two procedures depending on the stage of pregnancy: chorionic villi sampling (CVS) or amniocentesis. The details of each procedure are tabulated below.

Collection procedure

Guided by ultrasound, an OB-GYN uses a catheter through the vagina or a long, hollow needle through the abdomen (depending on the position of the fetus) to collect cells.

Viability Test
A DNA viability test is performed on unusual samples to check for the presence of usable DNA for a paternity or other DNA tests. In paternity cases where the alleged father is missing or unavailable, we may be able to use preserved samples such as blood, hair, or other body tissues.
The viability test fee is variable and depends on sample type and condition. This non-refundable fee is separate from the paternity test fee and needs to be paid before testing begins.
